Development of Analytical Method for Estimation of Curcumin Demethoxy Curcumin and Bis Demethoxy Curcumin from Fast Dissolving Tablet Containing Curcuma Extract

Curcumin, demethoxy curcumin and bis-demethoxy were isolated from the dried root powder of Curcuma longa through combined application flash chromatography open column chromatography. Isolated constituents identified using spectral methods. An analytical method was developed for simultaneous quantitative estimation curcuminoids fast dissolving tablets containing ethanolic extract sp., as an active ingredient. The separation achieved in a C18 isocratic mobile phase comprised acetonitrile:water (aqueous 4 % v/v glacial acetic acid) (50:50 v/v), with flow rate set at 2.0 ml/min. chromatogram recorded 425 nm, photodiode array detector. validated per international conference on harmonisation guidelines. found to be linear concentration range 5-120 μg/ml, precise robust minor modifications selected chromatographic parameters. average recovery within 98 %-102 %. Limit detection limit quantification value determined 0.35 1.06, respectively. 100 mg formulated directly compressible vehicle. Each tablet contain 20.77 %±0.22 w/w curcumin, 3.18 %±0.03 0.33 %±0.006 when estimated method.
